Abstract

Calotropis procera is a commonly used herb in Ayurvedic medicine. This review supports all updated information on its phytochemical and pharmacological activities, traditional uses and scientific approach. The plant extracts and its chemical marker or target molecule Arecoline have been widely used for the treatment of a large number of human ailments. The chemical entities of this plant has been used as an antidiabetic, blood pressure regulating activity, antiulceogenic, antioxidant activity, anticonvulsant activity, C.N.S. stimulant activity, oxytocic activity, antifertility, anthelmintic and antiviral activity etc. Scientifically proved activities are related with traditional concept. Scientific evidence exists with respect to their major and minor constituents.

Abstract

A reverse phase high performance liquid chromatographic method was developed for the simultaneous estimation of Cefadroxil (CEF) and Probenecid (PROB) in bulk and tablet dosage form. The separation was achieved by Princeton SPHER-100 C18 column of configuration was selected for further study. (Princeton SPHER-100 C₁₈, 250×4.6 mm, particle size 5 μm.). CEF is freely soluble in methanol and slightly soluble in acetonitrile, water and insoluble in ethanol. Probencid is freely soluble in methanol, slightly soluble in acetonitile, water. Initially methanol and acetronitrile in different ratios were tried and then acetonitril and phosphate buffer (PH 2.5 adjusted with orthophosphoric acid). It was found that acetonitril and phosphate buffer (PH 2.5 adjusted with orthophosphoric acid). In the ratio 60:40 v/v with the flow rate of 1mL/min gives good resolution of peaks with minimum tailing as compared to other mobile phases. Detection was carried out at 242 nm. Retention time of Cefadroxil and Probenecid was found to be 2.213 and 6.62 min, respectively. The method has been validated for linearity, accuracy and precision. Linearity for Cefadroxil was found in the range of 10- 60μg/ml and Probenecid was in the range of 10-60 μg/ml. The percentage recoveries obtained for Cefadroxil and Probenecid were found to be in range of 99.52-99.81 % and 99.50-99.53% respectively.

Abstract

Form fill seal (FFS) technology is an automated computer operated technology, to prepare sterile products. Mostly it is applicable for I.V. infusion bottles. In this process all steps such as formation of container, filling of container with content and sealing of container are performed sequentially, consistently and automatically in a closed sterile chamber of machine. This technology reduces the contamination rate below 0.1%. The reason behind FFS technology is to reduce the contamination during production in a closed sterile chamber of a machine. Thus there should be no personnel intervention to reduce the chances of contamination during the manufacturing of sterile products. Again it gives more production in very low operational cost with high assurance of sterility.

Abstract

The main objective of this investigation was to formulate controlled release dosage form by using citric acid cross linked cellulose based hydrogel. Hydrogels were prepared by cross liking the polymer HPMC-K4M by using suitable cross linking agent, citric acid. Due to cross linking reaction hydrogel gave good swelling as well as controlled release property to metoprolol succinate hydrogels. In this study 3² full factorial design, which was augmented for 2 central points was used by using Design-Expert 8.0.7.1. It gave 11 runs. In this design the concentration of citric acid (X1) and reaction time (X2) were selected as independent variables, while as drug content, swelling index and t₇₅ of % cumulative drug release were selected as dependent variables. From the results it can be concluded that the effect of citric acid concentration on drug content and t₇₅ of % CDR was nonsignificant. But the effect of citric acid concentration on swelling index was much significant, means as citric acid concentration increased; there was decrease in swelling index of hydrogel. Again effect of reaction time on drug content as well as on t₇₅ of % CDR was much significant, means as reaction time increased; there was increase in drug content as well as t₇₅ of %CDR of hydrogel. But the effect of reaction time on swelling index was nonsignificant. On the basis of all responses obtained, drug content, swelling index and t₇₅ of % CDR of run R1 are best. Drug content, swelling index and t₇₅ of % CDR of this run are 99.5%, 276.64% and 207.6 min. respectively. So optimized formulation was run R1, which gave the best in vitro release up to 94.24% in 360 min (6hrs).IR and DSC study shows no evidence on interaction between drug, polymers and other excipient. The in vitro data were fitted to different kinetic models.

Abstract

Mobocertinib is a kinase inhibitor targeted against human epidermal growth factor receptor (EGFR). It is used particularly in the action of non-small cell lung cancer (NSCLC) caused by exon 20 placing mutations in the EGFR gene, which are classically linked through a poorer prognosis (as compared to "classical" EGFR mutants causing NSCLC) and are linked with opposition to standard targeted EGFR inhibitors. Mobocertinib appears to be an effective means of treating this otherwise treatment-resistant NSCLC, exerting an inhibitory effect on EGFR exon 20 insertion mutant variants at concentrations 1.5- to 10-fold lower than those required to inhibit wild-type EGFR.2 Mobocertinib, below the brand name Exkivity (Takeda Pharmaceuticals Inc.), was granted accelerated agreement by the FDA in September 2021 for the action of locally advanced or metastatic NSCLC in patients with EGFR exon 20 insertion mutations who have failed previous therapies.1
